TweakXP isn't quite as helpful here as I thought it might be. I'm guessing the registry keys involved are:

For global settings:

HKEY_LOCAL_MACHINE\SOFTWARE\Microsoft\Windows\CurrentVersion\Explorer\AutoplayHandlers

For per user settings:

HKEY_CURRENT_USER\Software\Microsoft\Windows\CurrentVersion\Explorer\AutoplayHandlers

Further information greatly appriciated

You might also look at HKEY_LOCAL_MACHINE\SYSTEM\CurrentControlSet\Services\Cdrom key
the value of 1 turns it on 0 is off
